If your host put you in the Tower Suites it has its own lobby and check in which is considered VIP check in. If you are RFB there is a special lounge with check in immediately on your right when you enter the Tower Suites lobby. Best way to be sure is just email and ask your host. Have fun!
Wynn's true VIP check-in used to be located in a room off of the main tower suites lobby to the right of the main tower suites check-in desk - but from what I understand they've redone the whole tower suites lobby since the last time I was there so it may have moved as part of the renovation.
Is your host sending a limo for you? If so, you're probably staying in the Tower Suites and they'll more than likely give the heads up of your arrival and walk you to where you need to go.
What MikeE said is correct. As you step out of the limo, your host or assigned bellhop will direct you where to go. Tower suites lobby has been redesigned but the VIP room is in the same location.

I've always checked in at the TS lobby even when on RFB. Never set foot in the RFB lounge. First couple of times I was on RFB, I didn't even know it existed! These days only get RFL so probably not welcome there anyway!
