If you stay at a hotel that has a shuttle from the airport, (several have the shuttle) you can then take a taxi. Crown Plaza I believe it is has a shuttle...Give me a bit more info and I can try to help.

I haven't done it before but, there is bus service from the airport. Looks like the AF goes downtown to Market Street Station, $17 r/t. Depending on where you are staying, you may be able to walk from there or catch the 16th Street Mall free ride to get you closer to your destination.

Skyride is the bus service. I have used it three times in the last three years, and it's worked great to get from DIA to downtown Denver and back. Check out the Skyride website here: http://www.rtd-denver.com/skyRide_SubHome.shtml
The previous poster was correct in noting that route AF is probably the one you want.
The bus can drop you off at Market Street Station (along with a few other stops close to downtown) and from there you can hop on the 16th street mall free shuttle. This might be unnecessary for you, it really depends on where in downtown Denver you're wanting to go.

I recommend buying your bus tickets online via the website above prior to your trip to denver. It'll save you a couple bucks, and you won't have to worry about having exact change to pay the bus driver when getting on the bus.

The buses are nice charter-style buses, with plenty of room to store your luggage underneath the bus (the bus operator will store and retrieve your luggage for you).
It's about a 45 minute ride from DIA to downtown Denver on this bus.

Thanks for all the info. I'm going to Vegas on Saturday and have a long layover in Denver and was told that I should go downtown to kill some time.
    Again, thanks!

Lots to do Downtown, 16th Street is main drag and as mentioned, frequent free buses run up and down. http://www.downtowndenver.com/

And if you want to get in mood for Sin City, have a beer at Tilted Kilt on 16th,
